酸胺缩合最理想方式:不用过柱子就可以拿到很纯的产物

2013年,来自英国伦敦大学学院的Tom D. Sheppard团队报道了一种新型Lewis酸B(OCH2CF3)3作为媒介的酸胺缩合反应,在绝大部分的合成例子中,最终产物都不需要柱纯化,通过商业可得的树脂经过简单的过滤即可得到纯品,不需要水层处理和柱层析。
该反应体系的底物范围非常之广,简单的酸到手性酸,苄胺与DMF的甲酰化反应等等都能顺利进行,以良好的产率和手性保持得到目标产物。反应体系往往只需要固相操作就可以得到纯度较高的产物。具体固相纯化方式见下图。
固相操作过程非常简单,具体如下所示(以下操作来自本文参考的文献):
Solid Phase Workup
After the indicated time, the reaction mixture was diluted with CH2Cl2or EtOAc (3 mL) and water (0.5 mL). Amberlyst A-26(OH) (150 mg), Amberlyst 15 (150 mg) and Amberlite IRA743 (150 mg) were added to the reaction mixture, and it was stirred for 30 min. MgSO4 was added to the reaction mixture, which was thenfiltered, the solids were washed three times with CH2Cl2or EtOAc, and thefiltrate was concentrated in vacuo to yield the amide product.
