Protective effect of curcumin against myocardium injury in ischemia reperfusion rats

背景与目的

长期以来,姜黄素是一种全球性的调味品和传统中药。目前研究探讨姜黄素对大鼠缺血再灌注心肌损伤的保护作用。

方  法

我们用姜黄素(剂量为10、20或30mg / kg / d)或盐水喂养SD大鼠,每组10只。二十天后,通过结扎左前降支冠状动脉60分钟后,再灌注心肌3小时,来制备大鼠心肌损伤的模型。然后,检测心肌组织中的脂质分布、脂质过氧化产物、抗氧化酶和基因表达。

结  果

喂养姜黄素组与对照组相比,氧化应激降低3倍,梗死面积减少2.5倍,有统计学意义;与缺血再灌注心肌损伤组(I / R)相比,大鼠梗死面积的百分比降低(从49.1%降至18.3%)。姜黄素可进一步增强STAT3的磷酸化,与剂量依赖有关。

结  论

姜黄素摄入可能通过激活JAK2 / STAT3信号通路,减少氧化应激损伤和抑制心肌细胞凋亡来降低冠心病的风险。

BACKGROUND:Curcumin has long been used as a condiment and a traditional medicine worldwide.The current study investigates the possible protective effect of curcumin on heart function in myocardium ischemia-reperfusion (MIR) rats.

METHODS:  We fed Sprague–Dawley (SD) rats (10 in each group) either curcumin (10, 20 or30 mg/kg/d) or saline. Twenty days later, the rats were subjected to myocardial injuries by ligating the left anterior descending coronary artery (60 min), and subsequently, the heart (3 h) reperfused by releasing the ligation. Then, lipid profile, lipid peroxidation products, antioxidant enzymes and gene expression were assessed in myocardium tissue.

RESULTS: Only the rats that were supplemented with curcumin (10, 20 or 30 mg/kg/d) showed significant (p < 0.05) reductions in oxidative stress (3-fold), infarct size (2.5-fold), which was smaller than that of the control group. The percentage of infarct size in MIR rats with curcumin at 10, 20 or 30 mg/kg/d decreased(from 49.1% to 18.3%) compared to ischemia-reperfusion (I/R). The enhanced phosphorylation of STAT3 was further strengthened by curcumin (10, 20 or 30 mg/kg/d) in a dose-dependent manner.

CONCLUSIONS: Curcumin intake might reduce the risk of coronary heart disease by stimulating JAK2/STAT3 signal pathway, decreasing oxidative damage and inhibiting myocardium apoptosis.
