NAD Precursor Supplementation Increases Lifespan in Mouse Models
Studies in mouse models have shown that supplementation with NMN can increase lifespan, decrease and age-related frailty. This is likely due to NAD being a coenzyme for the sirtuin family of proteins, which can activate cellular repair pathways and which have shown to be involved in increasing lifespan and healthspan.
“The observed correlation between NAD levels and aging adds weight to the idea that NAD may play a role in cell senescence and longevity”
“Maintenance of intracellular NAD may therefore be beneficial in counteracting at least some of the age-related cellular degenerative processes.”
NMN Supplementation and Type 2 Diabetic mice
Supplementing diabetic mice with NMN was able to ameliorate Type 2 Diabetes-associated pathophysiologies. Mice given NMN displayed increased insulin sensitivity, decreased blood sugar and overall improvements in metabolic health markers.
“NAD biosynthesis is compromised by HFD and aging, contributing to the pathogenesis of type 2 diabetes.”
“promoting NAD biosynthesis by administering NMN, a key NAD intermediate, can be an effective intervention to treat the pathophysiology of diet- and age-induced Type 2 Diabetes. “
NAD Supplementation and Alzheimer’s mouse models
Studies in Alzheimer’s model transgenic mice have shown significant improvement in symptoms with NMN treatment. Mice treated with NMN showed improved memory, cognitive function as well as a decrease in beta-amyloid plaques in brain tissue.
“The data prove that NMN treatment greatly reduces multiple Alzheimer’s-disease-associated pathological characteristics,”
“In summary, this study provides essential preclinical evidences that NMN takes effects in reversing cognitive deficits and substantially lowering the burden of amyloid plaque, neuroinflammation, cerebral amyloid-β concentrations, and loss of synapse in middle-aged ADTg mice.”
NMN and Vascular Health in mice
Arterial stiffening is associated with advanced age and is a major contributor age-associated heart disease. In addition to being associated with heart problems, stiffening of arteries has been linked to cognitive impairments, metabolic disorders and age-related muscle loss. Treating old mice with NMN significantly improved arterial health and reduced arterial stiffness compared to untreated old mice.
“The results of this study provide the first evidence that oral supplementation with NMN represents a novel strategy for combating arterial aging and its pathological sequelae.”
