脂质体封装的AC-dots及其细胞摄取研究。由于AC-dots表面带负电荷,与细胞膜表面有排斥作用,因此文中通过阳离子脂质体将其封装以提高AC-dots的内化效率(A),从图B中的荧光成像结果能够明显看出脂质体封装后AC-dots能够被细胞高效内化。C-E图为不同浓度脂质体封装AC-dots标记的细胞的CEST MRI测量结果,证明脂质体不会影响AC-dots的CEST信号。

AC-dots标记的U-87 MG细胞移植到小鼠脑部1天后的CEST MRI。(B-D)图实验结果表明AC-dots能够增强CEST对比效果,但均匀度不高。同时AC-dots的荧光成像(E)结果与CEST MRI结果得到了相互印证。

该工作证明了亲水性碳点AC-dots具有明显的CEST MRI对比度,能够用于体内和体外的CEST MRI,这主要归因于粒子表面可交换的羟基和胍盐质子。这项工作将会在设计和合成具有新结构和功能的CEST碳点领域进一步扩展,使碳点作为CEST MRI造影剂在活体成像中重新焕发活力。
