液相分离在同位素标记代谢组学的应用

研究方向有:质谱蛋白质组学和代谢组学、阿尔茨海默病的分子发病机制、泛素和蛋白质组Turnover和癌症的系统生物学。

高通量非靶向代谢组学通常依赖于互补液相色谱-质谱(LC-MS)方法来扩大各种代谢物的覆盖范围,但这些方法的集成尚无法完全表征。我们系统地研究了8种LC-MS设置,不同固定相(HILIC和C18),流动相(酸性和碱性pH)下亲水相互作用液相色谱(HILIC)-MS和纳流反相液相色谱(nRPLC)-MS的性能)和MS电离模式(正负)。之前曾报道过nRPLC-MS的优化,但我们在HILIC-MS(2.1 mm×150 mm)中发现,通过从2 mg细胞/组织样本。由于峰特征受到污染物的高度损害,因此我们使用稳定同位素标记的酵母来增强配方鉴定,以比较不同的LC-MS条件。8种LC-MS设置可检测总共1050个配方,其中先佳组合分别为4种,3种和2种LC-MS可以回收78%,73%和62%的配方。此外,在存在或不存在氮饥饿的情况下收集这些酵母样品,从而能够对比较的配方和代谢物结构进行定量比较,然后使用选定的合成代谢物进行验证。

结果表明,氮饥饿会下调氨基酸成分,但会上调尿苷相关的代谢。
